Miyazato, who shot a first-round 63 to claim the lead in Morelia, overtook second-round leader Michelle Wie with a two-under-par round of 71 on Saturday.
Against this Wie, who is tied with fellow American Brittany Linciome (72) shot an even-par 73 to trail Miyazato by one with a round to play.
Stacy Lewis of the USA and Na Yeon-choi of South Korea are in tied fourth, followed by home favourite Lorena Ochoa in sixth.
The world number one, who will retire from professional golf after this event, carded a 70 which featured five birdies and two bogeys to remain in contention going into the final day..
